An air sample that pulls a measured volume of air across a sticky slide so a lab can count and identify the mold spores in it.
A spore trap cassette is placed on a calibrated pump for a set run time, so the lab knows exactly how much air passed over the collection surface. The lab then counts and identifies what it captured under a microscope and reports concentrations by genus. That yields a number, which is why spore traps are the most common air sampling method, and also why they are the most commonly over-interpreted.
The number only means something in comparison. There is no published safe threshold for airborne mold spores, and neither CDC nor EPA sets one, because the response varies enormously between individuals. What an assessor actually evaluates is the relationship between the indoor sample, an outdoor control taken the same day, and samples from unaffected areas of the same building. Indoor concentrations at or below the outdoor baseline with a similar mix of genera suggest a normal fungal ecology. Indoor concentrations dominated by genera that are scarce outdoors suggest an indoor source.
Both agencies are also clear that sampling is not the way to decide whether visible mold needs to be removed. If you can see growth and there is a water source, the sample adds cost without changing the answer. Sampling earns its place when growth is suspected but not visible, when the extent of contamination needs defining, or when a completed remediation needs verifying.
A useful sampling set has at least three samples: the affected area, an unaffected interior control, and an outdoor control, all taken the same day with the same equipment. A single indoor sample with no control is not interpretable, however precise the lab report looks.
Tape lifts and swabs answer the different and often more useful question of whether a particular surface has growth on it, and they are what confirms that a stain on a closet wall is mold rather than dust or water staining.
Under Florida law, performing mold assessment for compensation requires a state mold assessor license, and the assessor cannot be the company that remediated the structure within the previous twelve months. That separation is what makes a clearance result worth anything.
